Use | (3R)-3-(hydroxymethyl) tert-butyl morpholine-4-carboxylate is a key intermediate in the synthesis of BMS-599626, a selective and orally potent inhibitor of BMS-599626 human epidermal growth factor receptor 1 and 2 kinase. As an organic chemical intermediate, N-substituted morpholine has the dual properties of tertiary amine and ether, and can be used as a catalyst for the production of polyurethane foam, aminobenzylpenicillin and carboxy benzylpenicillin synthesis catalyst, but also can be used as surfactants, extractants, corrosion inhibitors, it is an ideal spinning solvent for the manufacture of Lyocell natural fiber and Newcell rayon filaments. |
preparation | N-substituted morpholine contains dual groups of tertiary amine and ether, it is not only an important catalyst in the process of semi-synthesis of penicillin and polyurethane foaming, but also an important raw material for the synthesis of N-substituted morpholine oxide. The current synthesis process mainly includes morpholine method, N-methyl diethanolamine method, diethanolamine method, diethylene glycol method and dichloroethyl ether method. (3R)-3-(hydroxymethyl) morpholine-4-carboxylic acid tert-butyl ester substituted with (R)-(4-benzyl-3-morpholinium)-Methanol as the starting material by reaction with di-tert-butyl dicarbonate to prepare [1], synthesis reaction formula as shown below: Figure 1 (3R)-3-(hydroxymethyl) reaction scheme for the synthesis of tert-butyl morpholine-4-carboxylate experimental operation: triethylamine was added to a solution of (R)-(4-benzyl-3-morpholinium)-methanol in dichloromethane, di-tert-butyl dicarbonate in dichloromethane was added dropwise and the reaction was completed for 15 hours. The solvent was distilled off under reduced pressure and the concentrate was chromatographed on silica gel column [eluent: V (ethyl acetate) /V (petroleum ether)]. = 1/10] the compound (3R)-3-(hydroxymethyl) morpholine-4-carboxylic acid tert-butyl ester was purified. |
